How it works

Sample to signal in one incubation

Add sample and HRP-modified anti-CD9/CD63/CD81 antibodies, incubate, wash, add substrate, and read. Intact EVs are captured and detected in a single step — no lysis or purification required.

The protocol

Minimum hands-on time

A single ~3-hour incubation and one wash — about 15 minutes of hands-on time from sample to readout.

Incubation

Add diluted sample and the antibody mix to the well plate and incubate (~3 hours).

Wash

Wash the wells 5× with Wash Buffer to remove unbound material.

Signal generation

Add 100 µL TMB substrate (5–10 min), then 100 µL Stop Solution.

Signal readout

Measure absorbance on a standard plate reader — or luminescence for the Lumi format.

What does the Atlas EV ELISA detect?

It uses antibodies against the tetraspanins CD9, CD63 and CD81 — detecting all three simultaneously, as both capture and detection antibodies — to quantify intact extracellular vesicles.

How long does the assay take?

There is a single ~3-hour incubation followed by one wash step, with only about 15 minutes of hands-on time. Then add substrate, stop, and read.

Do I need to purify my sample first?

No. The assay runs directly on biofluids (plasma, serum, urine, CSF, cell culture media) or on purified EVs such as SEC fractions, using only 50 µL — no purification or concentration required.

What is the limit of detection?

Yes — the Atlas Lumi Homebrew format supports customizable capture antibodies for a one-step custom assay. Just provide 100 µg of your antibody.

Do I have to run the whole plate at once?

No. The kit has 12 strips of 8 wells, so you can run individual strips or scale up to the full 96-well plate.
